摘要:
目的 探讨微小RNA(miRNA)-145在脑胶质瘤组织及胶质瘤细胞系中表达变化及对胶质瘤细胞系U251细胞增殖和凋亡的影响。方法 采用实时荧光定量PCR检测检测10例胶质瘤组织和瘤周组织以及4种细胞系(人脑胶质细胞株HEB以及胶质瘤细胞系U251、T98和U87)miRNA-145的表达水平。将miRNA-145模拟物、阴性对照序列转染胶质瘤细胞系U251细胞,采用PCR检测U251细胞miRNA-145表达水平,采用MTT法检测U251细胞增殖能力,采用流式细胞仪检测U251细胞凋亡。结果 与瘤周组织相比,胶质瘤组织miRNA-145表达水平明显降低(P<0.05)。与正常胶质细胞HEB相比,胶质瘤细胞系U251、U87、T98细胞miRNA-145表达水平均明显降低(P<0.05)。与阴性对照组相比,miRNA模拟物组U251细胞miRNA-145表达水平明显增高(P<0.01),U251细胞增殖能力明显降低(P<0.05),U251细胞凋亡水平明显增高(P<0.05)。结论 胶质瘤组织miRNA-145呈低表达,上调miRNA-145表达能够抑制胶质瘤细胞系U251细胞增殖能力并诱导U251细胞凋亡。这提示miRNA-145有可能成为胶质瘤治疗的靶点。
Abstract:
Objective To investigate the effects of miRNA-145 on the proliferation and apoptosis of U251 glioma cells. Methods Quantitative real-time reverse transcription polymerase chain reaction (qRT-PCR) was applied to detect the expression of miRNA-145 in 10 human peritumoral tissues and 10 glioma tissues, and 3 glioma cell lines and normal glial cells. The miRNA-145 mimics were transfected into U251 glioma cells. The proliferation and apoptosisof U251 glioma cells was determined by Mosmann Tetrazolium test (MTT) assay and flow cytometry (FCM), respectively. Results The level of miRNA-145 expression in the glioma tissueswas significantly lower than that in the peritumoral tissues (P<0.05). The expression levels of miRNA-145 in glioma cells of U251, U87 and T98 were significantly lower than normal glial cells line HEB (P<0.01). The transfection of miRNA-145 mimics into U251 glioma cells significantly increased the expression of miRNA-145 in U251 glioma cells, decreased U251 cell proliferation and induced U251 cell apoptosis (P<0.05). Conclusions The expression of miRNA-145 is significantly decreased in glioma tissues and U251 cells. The up-regulation of miRNA-145 can inhibit the proliferation and induce apoptosis of U251 cells. The miRNA-145 might be a new molecular target for the treatment of glioma.
