摘要:
目的 探讨醒脑静注射液对大鼠蛛网膜下腔出血(SAH)后早期脑损伤(EBI)的影响。方法 将36只成年雄性SD大鼠随机分为对照组、模型组、醒脑静组,各12只。颈内动脉血管刺破法制备SAH模型,对照组不刺破血管而其余操作同模型组,醒脑静组造模成功后腹腔注射醒脑静注射液[10 ml/(kg.d)],模型组和对照组给予同等量生理盐水。造模后24 h,Garcia JH评分评估大鼠神经功能,干湿重法评估脑水含量,ELISA检测海马组织IL-1β及IL-18水平,免疫组化染色和免疫印迹法检测海马组织NLRP3及Caspase-1的表达水平。结果 与对照组比较,模型组大鼠Garcia JH评分明显降低(P<0.05)、脑水含量明显升高(P<0.05),海马组织IL-1β、IL-18、NLRP3、Caspase-1水平明显升高(P<0.05)。与模型组比较,醒脑静组Garcia JH评分显著增高(P<0.05)、脑水含量明显降低(P<0.05)、海马组织IL-1β、IL-18、NLRP3、Caspase-1水平明显降低(P<0.05)。结论 醒脑静注射液可改善大鼠SAH后EBI,可能与减轻神经细胞焦亡水平有关。
Abstract:
Objective To investigate the effects of Xingnaojing injection on early brain injury (EBI) in rats after subarachnoid hemorrhage (SAH). Methods Thirty-six adult male SD rats were randomly divided into control group, model group, and Xingnaojing group, with 12 rats in each group. The SAH model was established by puncturing the internal carotid artery. The control group did not undergo vascular puncture, while the rest of the procedures were the same as in the model group. After successful modeling, the rats in the Xingnaojing group received intraperitoneal injection of Xingnaojing injection (10 ml/kg/d), while the rats in the model and control groups received an equal amount of normal saline. At 24 hours post-modeling, neurological function of rats was evaluated using the Garcia JH score, brain water content was assessed using the wet-dry method, IL-1β and IL-18 levels in the hippocampal tissues were measured by ELISA, and the expression levels of NLRP3 and Caspase-1 in the hippocampal tissues were detected by immunohistochemistry and immunoblotting. Results Compared to the control group, the rats in the model group showed a significant decrease in Garcia JH score (P<0.05), a significant increase in brain water content (P<0.05), and a significant increase in IL-1β, IL-18, NLRP3, and Caspase-1 levels in the hippocampal tissues (P<0.05). Compared to the model group, the rats in the Xingnaojing group had a significant increase in Garcia JH score (P<0.05), a significant decrease in brain water content (P<0.05), and a significant decrease in IL-1β, IL-18, NLRP3, and Caspase-1 levels in the hippocampal tissues (P<0.05). Conclusion Xingnaojing injection can improve EBI in rats after SAH, possibly by reducing the level of neuronal pyroptosis.
