摘要:
目的 探讨姜黄素对人胶质瘤SHG44细胞侵袭和迁移的影响。方法 胶质瘤SHG44细胞使用含10%胎牛血清DMEM培养基培养,终浓度分别5、10、20、40 μmol/L姜黄素作用24、48、72 h;对照组加入等量二甲基亚砜。CCK-8比色法检测细胞增殖活力;细胞划痕实验检测细胞迁移距离及迁移率;Transwell实验细胞侵袭能力;免疫印迹法检测胶质瘤SHG44细胞基质金属蛋白酶(MMP)2、9表达。结果 姜黄素对SHG44细胞生长具有显著抑制,且呈时间与浓度依赖性(P<0.05);最佳作用浓度在10~20 μmol/L。与对照组相比,姜黄素作用后,SHG44细胞迁移距离、迁移率、侵袭能力明显降低(P<0.05),SHG44细胞MMP2、MMP9蛋白表达水平显著下降(P<0.05)。结论 姜黄素可抑制胶质瘤SHG44细胞的增殖,进而抑制细胞的迁移及侵袭能力,且呈时间及浓度依耐性,其机制可能与下调MMP2和MMP9蛋白表达有关。
Abstract:
Objective To study the effect of curcumin on invasion and migration of human glioma cell line SHG44 and its mechanism. Methods CCK-8 kit assay was used to assess the effects of curcumin (0, 5, 10, 20, 40μmol/L) on the proliferation of human glioma cell line SHG44 24, 48, 72 hours after the action of curcumin. The cell scratch test and Transwell assay were used to evaluate the effects of curcumin on the invasion and migration of SHG44 cells. Western blotting was used to detect the expressions of matrix metalloproteinase (MMP-2) and MMP-9 in SHG 44 cells. Results CCK-8 kit assay showed that the curcumin significantly inhibited the proliferation of SHG44 with a concentration and time-dependent manner. The invasion and migration of SHG44 cells exposed to curcumin were significantly suppressed compared with the control group (P<0.05). The curcumin significantly reduced the expression levels of MMP-2 and MMP-9 in SHG 44 cells compared with the control group (P<0.05). Conclusions Curcumin can inhibit the proliferation, invasion and migration of human glioma cell line SHG44 with a concentration and time-dependent manner. The mechanism may involve in the down regulation of MMP-2 and MMP-9 in SHG 44 cells.
