摘要:
目的 探讨姜黄素对人脑胶质瘤U251细胞趋化运动、侵袭能力的影响及其机制。方法 U251细胞置于含10%牛血清白蛋白的RPM1640培养液中培养,用不同浓度姜黄素作用U251细胞,用免疫组化染色并在激光共聚焦显微镜下观察U251细胞环氧化酶2(COX-2)蛋白表达,Transwell小室实验判断姜黄素对U251细胞运动、侵袭能力的影响。结果 姜黄素对U251细胞的生长具有抑制作用,而且呈浓度-时间依赖性(P<0.05)。姜黄素对U251细胞COX-2蛋白表达具有明显抑制作用(P<0.05),姜黄素对U251细胞运动、侵袭能力具有明显抑制作用(P<0.05)。结论 姜黄素可能通过抑制U251细胞COX-2蛋白表达从而抑制细胞的趋化运动及侵袭能力;并呈时间-浓度依赖性。
Abstract:
Objective To study the effect of curcumin on the migration and invasion of human glioma cells and its mechanism. Methods The expression of cyclooxygenase-2 (COX-2) protein was determined by immunohistochemical technique in U251 glioma cells which were treated with curcumin of different concentration. The abilities of migration and invasion were determined by transwell test in U251 cells treated by curcumin of different concentration. Results The expression level of COX-2 protein in U251 glioma cells significantly decreased by curcumin in a concentration and time-dependent manner and was negatively related to the concentration and curcumin treatment duration (P<0.05). Curcumin inhibited the abilities of the migration and invasion of U251 glioma cells in concentration and time-dependent manner. The abilities of the migration and invasion of U251 glioma cells were positively related to the levels of COX-2 protein expression. Conclusion That curcumin inhibits the abilities of the migration and invasion of U251 glioma cells in dose and time-dependent manner may be produced via inhibition of expression of COX-2
